Figure 1:This figure shows a rat
Purkinje cell in primary culture. Subsequent to loading with Fura-2AM
(3 uM for 25 min in 5% CO2 at 36 degrees C.), the culture was
equilibrated at room temperature for 35 minutes in artificial
CSF. The culture was then utilized for calcium imaging. Relative
intensity of intracellular calcium is indicated on the vertical
scale bar, with red to white indicating the highest concentration
(approximately 1 uM). The horizontal scale bar=50 um. Used with

magnification of the extensive, probably axonal, process in Figure
1. This frame is taken subsequent to glutamate administration
(10 uM). Note red calcium "hot spots" along the process,
in response to glutamate-induced depolarization. (bottom) Same
process, 50 msec later. Note disappearance of "hot spots".
